B. A. C. B. - Animals affiliate with same-species (conspecific) individuals for better survival, but particular affiliation patterns vary with age. - For the freshwater cichlid Astatotilapia burtoni, adult social behaviors are well studied in labs but juvenile social affiliations are largely unknown. - In related fish, juveniles (fry) recognize visual and chemosensory social cues of parents or siblings (kin)2. Do burtoni display this behavior? - Characterizing early social behaviors in model vertebrates aids understanding of social development and treatment approaches for neurodevelopmental and behavioral disorders. Social Affiliation Supports Survival Throughout Development Research Questions Two-Choice Assay for Social Cue Preference An Age Onset for Maternal Preference? - Overall, fry showed no clear preference for their mother over an unrelated male. - Older fry spent more time close to their mother. - At 10 dpf, fry may have not yet developed enough to recognize threats or their protective mother. - There may be an onset of maternal preference coinciding with release age. Future Directions - Experimentally redetermine fry release timeline by video tracking mouthbrooders (lab population’s behavior may drift from literature). - Test maternal and social preference at a wider age series, evaluate validity of proposed model - Investigate specific roles of vision and olfaction in early social cue recognition. - Identify candidate hormones and brain region(s) involved in early social affiliation. B) Using 1 fry as a focal animal, we tested preference for a group of 5 kin vs i) 1 kin. ii) 5 non-kin conspecific fry. iii) 5 heterospecific cichlid fry. 1. Place focal fry and two stimulus groups in a two-choice preference arena. 2. Take a 60-minute overhead video of uninterrupted behavior. 3. Track focal fry movement with idtracker.ai software to quantify time spent in either preference “zone”. Figure 3. Mother vs Unrelated Male burtoni. A) Friedman test revealed no significant difference in time spent between zones, p = 1.0 (n = 4 trials, age 10 & 13 dpf). B) Maternal preference increased in the 13 dpf group but was not statistically significant (p = 0.1199, two sample t-test). Figure 4. A) 5 Kin vs 1 Kin. Friedman test revealed no significant difference in time spent between zones, p = 0.1353* (n = 2 trials, age 13 dpf). B) 5 Kin vs 5 Non-Kin Conspecifics. Friedman test revealed no significant difference in time spent between zones, p = 0.8465 (n = 6 trials, age 13-15 dpf). C) 5 Kin vs 5 Heterospecifics. Friedman test revealed no significant difference in time spent between zones, p = 0.6065 (n = 6 trials, age 15-16 dpf). *Using a preference index, preference for 5 kin over 1 kin was statistically significant (p = 0.0179, one sample t-test).
